摘要
目的:对胃癌与萎缩性胃炎胃黏膜双向电泳(2-DE)图谱进行比较分析,筛选差异表达蛋白质。方法:利用双向凝胶电泳(two-dimensionalgel electrophoresis,2-DE)分离胃癌组和萎缩性胃炎组总蛋白质,并通过优化的计算机图像分析软件分析电泳图谱。结果:在胃癌组织中点检测共有蛋白质点1 137个,萎缩性胃炎组织中点检测共有蛋白质点1 096个,发现两者间共有104个差异蛋白质点,其中40个点仅在胃癌组织中表达,21个点仅在萎缩性胃炎中表达,33个点在胃癌组织中高表达,47个点在胃癌组织中低表达。结论:通过优化的双向凝胶电泳和图像分析技术对胃癌及萎缩性胃炎蛋白质组进行研究,获得了胃癌蛋白质组与萎缩性胃炎组的差异蛋白质点,为进一步利用质谱分析技术做差异蛋白质鉴定奠定了基础。
Objective To analysis the two-dimensional electrophoresis (2-DE) maps of gastric cancer and atrophic gastritis and to screen differently expressed proteins between them. Methods 2-DE (two- dimensional gel electrophoresis) was used to sever the general protein from mucosa tissue in gastric cancer and atrophic gastritis gastric, then analysed the results by computer-graph analysis software. Results We verifed 1137 protein spots in gastric cancer tissue and 1096 ones in atrophic gastritis specimens. Among them, there were 104 discrepancy protein spots. We found that 40 spots only emerged in gastric cancer and 21 ones in atrophic gastritis mucous. Meanwhile, 33 spots frenquently emerge and 47 ones were less expressed in gastric cancer tissue. Conclusion We got the discrepancy protein spots and laid hard basis for futther research of discrepancy protein.
