摘要
目的:探讨缺氧诱导因子-1α(HIF-1α)和CD105蛋白表达情况与骨肉瘤生物学特性的关系及临床意义。方法:采用免疫组化SP法检测36例骨肉瘤组织中HIF-1α和CD105的表达情况,并分析与临床病理参数及预后的关系。结果:HIF-1α表达水平和CD105-MVD与骨肉瘤的分化程度密切相关(P<0.01),与性别、年龄、肿瘤体积及临床分期无明显关系(P>0.05)。HIF-1α阳性表达组的CD105-MVD(35.28±4.91)显著高于阴性表达组(30.01±5.26,P<0.01),且HIF-1α表达水平和CD105-MVD呈正相关(r=0.6035,P<0.05);有转移组病例的HIF-1α表达水平和CD105-MVD均高于无转移组病例(P<0.01);单因素和多因素生存分析显示HIF-1α、CD105高表达与患者预后不良有关(P<0.05)。结论:HIF-1α、CD105高表达可能在骨肉瘤的恶性进展中发挥重要作用,并与肿瘤血管形成密切相关,二者联合检测对评价骨肉瘤的预后有一定价值。
Objective:To investigate the expressions of hypoxia - inducible factor -1α and CD105 in osteosarco- ma and their relationship with biological characteristics and clinical significance. Methods: The expressions of HIF -1α and CD105 were examined by SP immunohistochemical staining in 36 case of osteosarcoma tissues, and their re- lation to the clinicopathologic parameters and prognosis was analyzed. Results: The expressions of HIF - !ot and CD105 were closely associated with differentiation degree of the tumors (P 〈 O. 01 ), had no obvious relationship to sex,age,size of tumor and clinical stage(P 〉0.05). The CD105 -MVD value (35.28 ±4.91 )in HIF- lot positive group was significantly higher than those(30.01 ± 5.26) in negative group (P 〈 O. O1 ), and there was significant positive correlation between expressions of HIF- 1 ot and CD105 -MVD( r = 0. 6035, P 〈 0. 05 ). The level of HIF-1α expression and CD105 -MVD with tumor metastasis was higher than that without metastasis( P 〈 0. 01 ). Univari- ate and multivariate survival analysis indicated that high expressions of HIF - lot and CDI05 were correlated with poor prognosis(P 〈0.05). Conclusion: The overexpressions of HIF -1α and CD105 may play important role during the malignant progression of osteosarcoma and they were closely associated with tumor angiogenesis, combined detection of HIF -1α and CD105 is valuable in judging prognosis in osteosarcoma.
