香椿叶醇提物对体外蛋白质非酶糖基化的抑制作用 被引量:2

Inhibitory Effects of Ethanol Extract from Toona sinensis Leaves on the Formation of Protein Non-enzymatic Glycation in vitro

摘要 [目的]应用体外蛋白质非酶糖基化反应系统,研究香椿叶提取物对蛋白质非酶糖基化终末产物生成的抑制作用。[方法]在体外蛋白质非酶糖基化反应系统中,分别加入不同剂量的香椿叶提取物或阳性对照药氨基胍,在不同时间分别取孵育样品检测其荧光强度,并结合SDS-PAGE电泳结果,研究香椿叶提取物是否对蛋白质非酶糖基化有抑制作用。[结果]在该体外系统中,蛋白质非酶糖基化产物的生成与孵育时间呈正相关,香椿叶提取物浓度依赖性抑制蛋白糖基化终末产物的生成,其中,1.00g/L香椿叶提取物组抑制率均达80%以上。SDS-PAGE电泳结果表明,香椿叶提取物抑制了蛋白质非酶糖基化终末产物的生成。[结论]香椿叶提取物对体外蛋白质非酶糖基化终末产物的生成有一定的抑制作用。 [Objective] Using an in vitro protein non-enzymatic glycation system,the inhibitive effects of ethanol extract of Toona sinensis leaves(TE) on protein non-enzymatic glycation end products formation were studied.[Method] In in vitro protein non-enzymatic glycation system,different doses of TE or positive control drug,aminoguanidine,were added respectively.At different times samples were taken and measured by fluorescence spectrophotometer.Also,SDS-PAGE assay was carried out to identify the effects of extracts.[Result] In vitro system,protein non-enzymatic glycation products of the generation had positive correlation with and incubation time,Toona sinensis leaf extract concentration-dependent inhibition of protein glycation end-products was generated, which,1.00 g/L of Toona sinensis leaf extract cells inhibition rate was higher than 80%.SDS-PAGE electrophoresis results showed that Toona sinensis leaf extract inhibited the formation of advanced glycation end products.concentration of 1.0 g/L inhibitive rates were higher than 80%. [Conclusion] Toona sinensis leaf extract have a certain degree of inhibition on formation of advanced glycation end products in vitro.
