摘要
目的:探讨雌激素对慢性脑缺血大鼠海马神经元的保护作用.方法:采用永久结扎去势大鼠双侧颈总动脉的方法建立慢性脑缺血模型,项背部皮下注射17β-雌二醇.硫堇染色观察海马神经元的形态变化,SP免疫组织化学法观察海马神经元胆碱乙酰基转移酶(ChAT)、细胞凋亡抑制蛋白Bcl-2和细胞凋亡促进蛋白Bax的表达.结果:模型组大鼠海马神经元排列松散,大小不规则,部分细胞固缩深染,核固缩.雌激素治疗组海马神经元细胞形态、大小与正常接近,偶见个别神经细胞胞核固缩,胞质浓染.与模型组同时间点相比,雌激素治疗组各时间点海马神经元Bcl-2和ChAT蛋白的表达升高,Bax蛋白的表达降低,且均呈时间依赖性.结论:雌激素可通过抑制海马神经元凋亡、上调海马神经元ChAT的表达,发挥对慢性脑缺血大鼠海马神经元的保护作用.
Objective: This study will initially investigate the protective effects of estrogen on the hippocampus neurons of chronic cerebral ischemia rats. Methods: A chronic cerebral ischemia rat model was established by permanently ligating the common carotid arteries on both sides of castrated rats, with subcutaneous iniection of estrogen at nucha and back. Thionine staining was used to observe the morphological changes of hippocampus neurons. Immunohistochemistry staining was used to observe dynamic changes of expression of bcl-2, bax and ChAT in the hippocampus neurons, and MiVnt image analytical system was used to quantitatively analyze the results. Results: In the model group, the neurons in each region of the hippocampus was loosely arranged, the size of neurons was irregular, and some neurons were pyknosis and deeply stained. In the estrogen group, the morphology and size of neurons in the hippocampus nearly reached normal, and several neurons could be seen pyknosis of nuclei and deep stainning of cytoplasm. Compared with the model rats at the same time point, the bcl-2 and the ChAT expression in the hippocampus neurons in the estrogen group were significantly higher, and the bax expression in the hippocampus neurons in the estrogen group were remarkably lower in a time-dependent manner. Conclusion: Estrogen can protect the hippoeampus neurons of chronic cerebral ischemia rats by inhibiting apoptosis of the hippocampus neurons and increasing ChAT expression of hippocampus cholinergic neurons.
