摘要
目的探讨抑肽酶对脑出血大鼠脑组织中水通道蛋白(aquaporin,AQP)-4、9(AQP-4、9)表达的影响。方法将SD大鼠随机分为假手术组、模型组、抑肽酶组,建立大鼠脑出血模型,测定脑组织含水量,采用RT-PCR法检测脑组织中AQP-4、9mRNA表达的变化;Western blot法检测脑组织中AQP-4、9蛋白含量的变化。结果 RT-PCR结果显示假手术组AQP-4、9mRNA含量最低(0.36±0.14、0.42±0.11)(P<0.05),抑肽酶组AQP-4、9mRNA含量(0.54±0.15、0.67±0.27)低于模型组(0.72±0.34、0.93±0.03)(P<0.05);Western blot结果显示模型组AQP-4、9蛋白在脑组织中的表达量(202.78±21.18、243.11±12.48)明显高于假手术组(134.33±10.21、101.12±9.45)和抑肽酶组(177.41±9.07、204.56±16.43),而抑肽酶组AQP-4、9蛋白表达量明显高于假手术组(P<0.05);假手术组脑组织含水量最低[(61.23±0.36)%](P<0.05),而模型组脑组织含水量最高[(86.98±1.06)%](P<0.05)。结论抑肽酶对脑出血大鼠的保护作用,可能是通过抑制AQP-4、9在脑组织中的表达,从而减轻脑水肿。
Objective To study the aquaporin-4,9 (AQP-4,9) effect of aprotinin on intracerebral hemorrhage in rats. Methods SD rats randomly divided into the sham operated group,model group and aprotinin group. Intracerebral hemorrhage models were established. Brain water content was measured,and the AQP-4,9 level was analyzed by semi-quantitative reverse transcription and polymerase chain reaction (RT-PCR) and Western blot. Results Among all the groups,the AQP-4,9 mRNA of sham operated group showed the lowest level (0.36±0.14,0.42±0.11)(P〈0.05). Compared with model group (0.72±0.34,0.93±0.03),the AQP-4,9 mRNA expression in aprotinin group (0.54±0.15,0.67±0.27) were decreased (P〈0.05). The AQP-4,9 albumen expression in model group (202.78±21.18,243.11±12.48) were highest than sham operated group (134.33±10.21,101.12±9.45) and aprotinin group (177.41±9.07,204.56±16.43),but the AQP-4,9 albumen expression in aprotinin group were higher than sham operated group. The water content of brain tissue in sham operated group (61.23±0.36) was lowest (P〈0.05),but model group (86.98±1.06) was highest in three groups (P〈0.05). Conclusions The aprotinin protects intracerebral hemorrhage by refrain the cerebral levels of AQP-4,9 and reduce the brain edema.
