摘要
目的探讨金黄色葡萄球菌(SAU)对人脐静脉内皮细胞(HUVEC)的黏附、内化及诱导细胞凋亡的发生、发展的机制和致病意义。方法常规方法制备SAU悬液,分别感染HUVEC细胞30 min、1、24、h。采用光学显微镜和荧光显微镜观察SAU对HUVEC的黏附、内化情况,采用流式细胞仪、DAPI荧光染色及DNA琼脂糖凝胶电泳分析SAU对HUVEC细胞诱导凋亡的影响。结果SAU与HUVEC共育30 min发现有细菌黏附细胞表面,1 h后有少量细菌侵入胞内,并伴有凋亡现象的发生,随着时间的延长,内化的细菌数没有增加,凋亡现象通过流式细胞仪、DAPI荧光染色及DNA琼脂糖凝胶电泳分析明显的被观测到。结论SAU通过黏附、内化侵入内皮细胞并诱导内皮细胞发生凋亡。
OBJECTIVE To investigate the adherence,internalization of Staphylococcus aureus by cultured human umbilical vein endothelial cells(HUVEC) and induce the apoptosis occurrence,development mechanism,pathogenic significance in endothelial cells.METHODS The S.aureus bacteria suspension were prepared by conventional method.The bacterial inoculum with HUVEC was incubated for 30 minutes,1 hour,2 hours and 4 hours,respectively.The adherence and internalization of S.aureus wereobserved by light microscope and fluorescence microscope.The influence on induced apoptosis in HUVEC cells was analyzed by flow cytometry assay,DAPI fluorescent staining and DNA agarose gel electrophoresis.RESULTS The adherence to cellular surface happened after incubated for 30min and a few bacteria invaded into cells with cell apoptosis after 1 hour.The numbers for internalization of S.aureus did not increase as time extended.But apoptosis occurrences were obvious by flow cytometry assay,DAPI fluorescent staining and DNA agarose gel electrophoresis.CONCLUSIONS S.aureus can induce apoptosis of endothelial cells by adherence,internalization and invasion into endothelial cells.
