我国间日疟原虫混合流行区分离株Dufry抗原结合蛋白基因多态性分析 被引量:2

Analysis of Genetic Polymorphism in Duffy-Binding Protein among Plasmodium vivax Isolates in Chinese Mixed Epidemic Area

摘要 为了解我国季节混合流行区云南省间日疟原虫红内期候选抗原Dufly抗原结合蛋白(DBP)基因多态性特点,提取19株我国云南株疟原虫基因组DNA,PCR扩增DBPRⅡ区基因片段。以SalI标准株作为参照,对所获得的基因序列进行排序分析,检测基因突变位点,以中性检验分析评估序列多态性。结果表明,中国间日疟原虫混合流行区云南分离株PvDBPRII的基因多态性与南美、中非等流行突变位点一致,未发现地区特异性的突变位点,但与国内单一流行区湖北和浙江地区突变位点存在一定差异。原因可能为整体上受到自然选择的影响,呈正向选择趋势。上述结果可为以PvDBPRⅡ为基础建立有效传播阻断疫苗的研究提供一定的线索。 To investigate the genetic polymorphism of region II in Duffy-Binding Protein of Plasmodium vivax (PvDBP R II ) in Yunnan province, China, P. vivax genomic DNA was extracted from Nineteen Yunnan samples, from which PvDBP R II was amplified with a polymerase chain reaction. The PCR products were directly sequenced, then alignments were done to find variant sites, genetic polymorphism were analyzed with the neutral theory. Compared with PvDBP R II from South America and Middle East, variant sites of Chinese isolates were relatively limited. In China, there were some difference in variant sites among Yunnan and Zhejiang, Hubei isolates. The observed polymorphism might be trending with natural selection, positive selection may be occurring in PvDBP R II and favouring the fixtion of amino acid replacements in isolates from Yunnan.
