摘要
目的研究地锦草提取物对红色毛癣菌角鲨烯环氧化酶和β-(1,3)-D-葡聚糖合成酶的影响,探讨其抗真菌作用机制。方法药物干预红色毛癣菌后分离菌体,按试剂盒要求处理样品,利用酶联免疫法和荧光分光光度法,研究地锦草提取物对红色毛癣菌角鲨烯环氧化酶和β-(1,3)-D-葡聚糖合成酶活性的作用。结果地锦草提取物256μg.m l-1时,可显著降低红色毛癣菌中角鲨烯环氧化酶的活性(P<0.01);不同浓度的地锦草提取物,均可降低红色毛癣菌β-(1,3)-D-葡聚糖合成酶的活性(P<0.01)。结论地锦草提取物抗真菌机制可能与抑制角鲨烯环氧化酶和β-(1,3)-D-葡聚糖合成酶的活性有关。
Objective To study the effect of Euphorbia humifusa extract(EHE) on squalene epoxidase and β- (1,3) -D- glucan synthase of T. rubrum and its meehanism. Methods Treated T. rubrum with EHE, studied the effects of EHE on squalenee epoxidase activity in T. rubrum cell membrane by ELISA test and β- 1,3 - gluean synthase activity in T. rubrum cell wall by fluorospectrophotometry. Results The ELISA test results showed that squalene epoxidase activity was decreased significantly (P 〈 0.01, vs growth control) in T. rubrum when the eoncentration of EHE was 256 μg · ml^ -1. The fluorospectrophotometry test results showed that β - ( 1,3 ) - D - gluean synthase activity was decreased significantly in T. rubrum treated with low, middle and high concentrations of EHE ( P 〈 0.01, vs growth eontrol). Conclusion The antifungal meehanism may be related to its inhibition on squalene epoxidase and β - (1,3) -D- glucan synthase activity in fungus.
