摘要
目的 探讨直肠癌组织中碳酸酐酶Ⅸ(CA-Ⅸ)和血管内皮生长因子(VEGF)的表达及与临床病理的关系.方法 应用蛋白免疫印迹方法 (Western blot)检测104例直肠癌、30例正常直肠组织、30例直肠腺瘤中CA-Ⅸ和VEGF的表达.结果 直肠癌组织中CA-Ⅸ和VEGF阳性表达率分别为72.1%和65.4%,高于正常直肠组织和腺瘤组织(P〈0.05).CA-Ⅸ的表达与Dukes'分期及淋巴结转移相关,Dukes'分期A至C期CA-Ⅸ阳性率下降(P〈0.05),淋巴结转移组CA-Ⅸ阳性率低于无淋巴结转移组(P〈0.05).直肠癌组织和系膜组织VEGF的表达与淋巴结转移关系密切,N2组VEGF阳性表达率分别为81.3%和75.0%,高于NO组的53.7%和25.9%(P〈0.05).CA-Ⅸ和癌组织和系膜组织的VEGF的表达无明显相关(r=0.092和r=-0.292).结论 CA-Ⅸ和VEGF表达与直肠癌发生、发展和浸润转移有关,但两者作用机制不同.
Objective To explore the expressions and role of carbonic anhydrases-Ⅸ (CA-Ⅸ) and vascular endothelial growth factor (VEGF) in rectal cancer. Methods The expression levels of CA-Ⅸ and VEGF in 104 pathologically diagnosed rectal cancer tissues, 30 normal tissues, and 30 cases of non-neoplastic rectal mucosa were detected by Western blotting. Results The overall positive expression rate of CA-Ⅸ and VEGF in rectal cancer was 72.1% and 65.4%, repeetively, which was higher than that in non-neoplastic tissue(P〈0.05). The expression of CA-Ⅸ was related to Dukes' stage and status of lymph node metastasis. CA-Ⅸ expression positive rate was decreased with the progression of the Dukes' stage from A to C. CA-Ⅸ expression in patients with lymph node negative involvement was higher than that in lymph node involvement, 85.2% vs 58.8%, 56.3%, respectly, with the difference being statistically significant,P〈0.05. The expression of VEGF was related to status of lymph node metastasis. VEGF expression in carcinoma tissues of patients with lymph node involvement (81.3%) was significandy higher than that in lymph node negative involvement (53.7%) ,χ^2=6.88, P〈0.05, and VEGF expression in mesorectal tissues with lymph node involvement (75.0%) was significantly higher than that in lymph node negative involvement (25.9%),χ^2=14.52, P〈0.05. There was no positive association between CA-Ⅸ and VEGF. Conclusion The hypoxia and angiogenesis are helpful to the judgment of prognosis.
