基于iTRAQ技术筛选肝细胞癌微血管侵犯的生物标记物 被引量:10

Screening of potential serum biomarkers of microvascular invasion in human hepatocellular carcinoma by iTRAQ technology

摘要 目的 利用核素标记的相对和绝对定量(iTRAQ)技术比较肝内无血管侵犯和微血管侵犯的肝细胞癌(HCC)患者的血清,筛选并鉴定其差异表达的蛋白质,以作为HCC微血管侵犯的生物标记物.方法 收集无血管侵犯和微血管侵犯的HCC患者血清各15例,组内等量混合后利用免疫亲和柱去除14种高丰度蛋白,iTRAQ试剂标记后的样本利用二维色谱分离,经串级质谱鉴定及相对定量.结果 鉴定得到75种蛋白质,其中差异表达的蛋白有20种(上调或下调20%),微血管侵犯组较无血管侵犯组上调的蛋白有12种,下调的有8种.结论 鉴定出多种与肝癌微血管侵犯相关的生物标记蛋白,为肝细胞癌转移和复发的预测和术后的干预治疗提供了实验依据. Objective To compare serum pmteome between patients with hepatocelhlar carcinoma (HCC) with or without mierovascular invasion utilizing isobaric stable isotope labeling (iTRAQ) technology, and screen and identifiy the differentially expressed proteins used as the potential biomarkers of microvascular invasion in HCC. Methods Two groups of sera were respectively collected from HCC patients with or without microvascular invasion (15 cases in each group). High-abundant proteins were depleted, iTRAQ technology combined with liquid chromatography tandem mass spectrometry (LC-MS/MS) was utilized to identify differentially expressed proteins between two groups. Results A total of 75 proteins with the confidential score above 95% were identified, in which 12 proteins up-regulated more than 1.2 fold and 8.0 proteins down-regulated more than 0.8 fold were considered as differentially expressed proteins between two groups. Conclusion Several serum biomarker proteins associated with microvascular invasion in HCC were identified in this study, which may provide important messages in prediction of HCC recurrence and metastasis and instructions for interventional therapy after operation, iTRAQ is one of the important proteomic techniques in dipping into tumor biomarker identification and molecular target discovery.
