运动对动脉粥样硬化小鼠细胞间黏附分子-1和血管细胞黏附分子-1表达的影响 被引量:3

Inhibiting effects of exercise on aortic adhesion molecule expression in atherogenesis in apolipoprotein E knockout mice

摘要 目的观察在动脉粥样硬化(AS)形成过程中,运动对实验小鼠主动脉细胞间黏附分子-1(ICAM-1)及血管细胞黏附分子-1(VCAM-1)表达的影响。方法选取8周龄ApoE基因敲除小鼠饲以“西方饮食”饲料建立AS模型,将其分为ApoE-安静组及ApoE^-/-运动组,ApoE^-/-运动组给予持续12周中等强度跑台运动;另同时选取c57BL/6J小鼠作为空白对照组(C57安静组),给予普通生长繁殖饲料喂养。于实验进行12周时采用免疫组化法检测各组小鼠主动脉ICAM-1及VCAM-1表达情况。结果C57安静组主动脉无ICAM-1、VCAM-1阳性表达,ApoE^-/-安静组小鼠主动脉可见大量ICAM-1、VCAM-1阳性表达,在AS早期阶段,以内皮层阳性表达较明显;在纤维斑块及粥样斑块阶段,阳性染色则主要分布于斑块深部脂质核心区,内皮层呈低水平表达,ICAM-1、VCAM-1阳性表达面积相近;运动可促使ApoE^-/-小鼠ICAM-1、VCAM-1阳性表达面积明显减少(P〈0.05)。结论运动能下调AS小鼠主动脉ICAM-1、VCAM-1表达,这可能是运动抗AS的重要机制之一。 Objective To investigate the effects of exercise on the expression of intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) in the aorta. Methods Eight-week-old apolipoprotein E (ApoE) knockout mice (ApoE^-/- ) were fed a " western diet" for 12 weeks to establish an atherosclerotic model, and C57 mice were used as a homologous group. They were divided into three groups: a C57 sedentary group (CC group, n = 15) , an ApoE ^-/- sedentary group (AC group, n = 15) and an ApoE^-/- exercise group (AE group, n = 15). Mice in the exercise group ran on a treadmill at 10 m/min for 30 min progressing up to 13 m/min for 60 rain gradually for 5 d/week for a total 12 weeks. At the end of experiments, immunohistochemical analysis revealed the expression of ICAM-1 and VCAM-1 in the aortas of the mice. Results Immunostaining revealed no expression of ICAM-1 or VCAM-1 in the mice of the CC group. However the immunostaining revealed ICAM-1 and VCAM-1 in the endothelia, lesions and media in the ApoE^-/- mice in the AC and AE groups. The expression was less intense in fibrous plaque and atheromatous plaque of the endothclium, but was pronounced at an early stage. Meanwhile ICAM-1 and VCAM-1 expression were detected in regions of the endothelium without lesions. It was noticed that exercise significantly decreased the relative area showing ICAM-1 or VCAM-1 expression. Conclusions Moderate intensity exercise for 12 weeks can decrease the expression of ICAM-1 and VCAM-1 in ApoE^-/- mice. This may he one of the anti-atherogenie mechanisms of exercise.
