Effect of Baicalein on Expression of Intercellular Adhesion Molecule-1 in Endothelial Cells Stimulated by Lipopolysaccharide

摘要 目的研究黄连解毒汤有效成分黄芩素对细菌脂多糖(LPS)诱导的人血管内皮细胞细胞间黏附分子-1(ICAM-1)和抑制蛋白κB(I-κB)表达的影响。方法分别用LPS(终浓度1μg/mL)或LPS+黄芩素(终浓度1μmol/L、10μmol/L、50μmol/L)处理生长良好的ECV-304细胞,用RT-PCR和WesternBlot法检测ICAM-1表达情况和I-κB含量变化。结果LPS刺激ECV-304细胞可促进I-κB降解,上调ICAM-1表达水平,与空白对照组比较有统计学意义(P<0.01);黄芩素预处理能抑制LPS诱导的I-κB降解,降低ICAM-1表达水平,与LPS组比较有统计学意义(P<0.05)。结论黄芩素可通过抑制I-κB降解,影响NF-κB炎症信号途径,下调ICAM-1的表达,这可能是黄连解毒汤抗动脉粥样硬化的作用机制之一。 Objective To investigate the effects of baicalein,an effective components of Huanglian Jiedu decoction,on expression of intercellular adhesion molecule - 1 (ICAM - 1) and inhibitor of kB (I - kB) in endothelial cells ECV - 304 induced by lipopolysaccha- ride(LPS). Methods ECV- 304 cells were cultured in vitro,and treated by different concentration of baicalein(1 μmol/L, 10 μmol/L, 50 μmol/L) with or without LPS(1 μmol/L). The mRNA and protein expression "of ICAM- 1 were determined by reverse tran- scription polymerase chain reaction(RT - PCR)and Western blot, respectively. The expression of I- kB was detected by Western blot. Results LPS up - regulated the expression of ICAM - 1 in endothelial cells significantly, and markedly inhibited the expression of I - kB com- pared with control group(P〈0.01). Following pretreatment with baicalein, LPS - induced ICAM - 1 expression was down - regulated in both mRNA and protein levels,and degradation of I - kB was inhibited significantly compared with LPS - treatment group(P〈20. 05). Conclusion Baicalein might effectively down regulated ICAM - 1 expression in LPS - induced endothelial cells through inhibiting the degradation of I - kβ. It might be associated with the amelioration of atherosclerosis damage by Huanglian jiedu decoction.
