不同用药方案对小鼠种植窗期子宫内膜白血病抑制因子及胞饮突表达的影响 被引量:11

Expression of LIF and pinopodes in mouse endometrium during the implantation window based on different treatment protocols

摘要 目的:探讨不同用药方案对小鼠种植窗期子宫内膜白血病抑制因子(LIF)和胞饮突表达的影响。方法:将12只去势雌鼠及36只雌性未孕动情期小鼠分为激素替代治疗(HRT)组、超排卵组、控制性超排卵(COH)组和对照组。采用免疫组化SP法检测小鼠种植窗期子宫内膜LIF蛋白的表达;RT-PCR法检测小鼠种植窗期子宫内膜LIF mRNA的表达;扫描电镜观察胞饮突表达的变化。结果:4组小鼠种植窗期子宫内膜LIF mRNA及蛋白的表达差异均有统计学意义(F分别为5.978,4.788,P均<0.05)。与对照组相比,HRT组、COH组和超排卵组LIF蛋白及mRNA表达均降低(P<0.01);HRT组高于COH组和超排卵组(P<0.01);COH组高于超排卵组(P<0.01)。HRT组小鼠种植窗期子宫内膜胞饮突的发育在形态上与对照组类似,时相上较对照组发育略延后;COH组胞饮突的发育形态上较对照组受到抑制,时相上延迟于对照组;超排卵组胞饮突的发育明显受到抑制,时相上远远滞后于对照组。结论:3种用药方案均可使子宫内膜LIF和胞饮突的表达受到影响,使内膜容受性受到损害;在对小鼠子宫内膜容受性的改善方面,HRT优于COH和超排卵,COH优于超排卵。 Aim:To evaluate the effects of different treatment protocols on the expression of LIF and pinopodes in mouse endometrium during the implantation window. Methods :A total of 12 ovariectomized mice and 36 female mice were randomly allocated into 4 groups:hormone replacement treatment(HRT) group, superovulation group, controlled ovarian hy- perstimulation(COH) group and control group,and uterine samples were collected on Day 4 and Day 5 of pregnancy. The expression of LIF protein was detected by immunohistochemistry. The LIF mRNA expression was measured by RT-PCR. Pi- nopodes were investigated by scanning electron microscopy (SEM). Results: There were significant differences in the ex- pressions of LIF protein and mRNA among the four groups ( F was 4. 788,5. 978, respectively, both P 〈 0.05 ). The expres- sions of LIF mRNA and protein in HRT group were higher than those in COH group and superovulation group(P 〈 O. 01 ) , and those in COH group were higher than in superovulation group( P 〈 O. O1 ). Compared with the control, the development of pinopodes in HRT group was similar,those in COH group lagged behind that of the control group and the development of pinopodes in superovulation group lagged behind obviously and was repressed. Conclusion:All the treatment protocols can affect the expression of LIF and pinopodes in the endometrium, and do harm to the uterine receptivity. HRT protocol is bet- ter than COH protocol and superovulation protocol, and COH protocol is better than superovulation protocol.
