NMDAR1在脑缺血模型大鼠不同时段的表达及针刺干预作用的研究 被引量:9

N-methyl-D-aspartate Receptor Expression in Cerebral Ischemic Rats at Different Time and Effect of Acupuncture Intervention

摘要 目的:探讨针刺对脑缺血模型大鼠N-甲基-D-天门冬氨酸受体(NMDAR1)亚单位NR1的干预作用。方法:采用热凝闭大鼠大脑中动脉致局灶性脑缺血模型(MCAO),观察Y迷宫实验大鼠的学习记忆能力及神经行为学评分,并采用免疫荧光法观察缺血2h、1d、3d后CA1区和CA3区NR1的表达规律及电针对其的影响。结果:在神经行为学评价方面,电针1d时段、3d时段比模型1d时段、3d时段评分均要低(P<0.05)。Y迷宫试验电针组与模型组比较时间明显减少,电针1d时段和3d时段与模型1d时段、3d时段比较,差异有非常显著性意义(P<0.01)。而NR1的积分光密度值总和(IOD值)在CA1区和CA3区的表达一致,在2h产生高表达,随后降低,电针组IOD值比模型组低(P<0.05)。结论:电针可通过调节NR1,降低NR1在脑缺血早期的表达,从而改善脑缺血模型大鼠脑组织的损伤。 Objective:To observe the effect of electro-acupuncture on regulating N-methyl-D-aspartate receptor(NMDAR1) expression in cerebral ischemic rats.Methods:Focal cerebral ischemia(FCI) rat models were established by middle cerebral artery occlusion(MCAO) with heat-coagulation method.The learning and memory abilities of rats as well as their neuroethological score were observed in Y maze test.Immunofluorescence method was used for the detection of NMDAR1 expression in CA1 and CA3 regions 2 hours,one day and 3 days after ischemia,and the effect of electro-acupuncture on NMDAR1 expression was also observed.Results:Neuroethological score was lower(P<0.05) and time for arriving the safe area in Y maze was shorter(P<0.01) in the electro-acupuncture group one day and 3 days after ischemia than those in the model group.The total index of optical density of NMDAR1 in CA1 region was similar to that in CA2 region.NMDAR1 expression was obvious 2 hours after ischemia,and then began to decrease,the decrease being obvious in the electro-acupuncture group than that in the control group(P<0.05).Conclusion:Electro-acupuncture can relieve the cerebral injury in cerebral ischemic rats through regulating NMDAR1 expression and decreasing its expression at early stage of cerebral ischemia.
