摘要
目的:观察大鼠局灶性脑梗死后大脑皮质RhoA和ROCK(Rho-associated kinases)的表达以及电针刺激对其表达的影响。方法:将成年健康雄性SD大鼠40只,随机分为正常组,假手术组,模型组和电针组,每组十只。模型组和电针组利用改进后的Longa方法制作大脑中动脉闭塞(middle cerebral artery occlusion,MCAO)模型,电针组在大鼠麻醉苏醒后90min进行电针刺激,每天一次,连续针刺14天。每组分别在1d,3d,7d,14d四个时间点进行神经行为学评分。14天时运用免疫组织化学法观测缺血区大脑皮质中RhoA和ROCK的分布和表达,利用免疫印迹法检测缺血区侧大脑皮质RhoA和ROCK的蛋白表达。结果:模型组大鼠神经行为学评分明显高于正常组和假手术组(p<0.01),14天后电针组的神经行为学评分明显低于同时段模型组(p<0.05);免疫组化结果显示,电针组的RhoA和ROCK表达较模型组明显减少(p﹤0.05);Western blot检测结果显示,电针组RhoA蛋白和ROCK蛋白的表达量较模型组RhoA蛋白和ROCK蛋白明显减少(p<0.05)结论:大鼠局灶性脑梗死后脑损区皮质RhoA与ROCK表明显上调,给予电针干预后可降低其表达,这可能是电针促进MCAO大鼠脑损区轴突再生的机制之一。
Objective:To observe the expression of RhoA and ROCK on MCAO ratS’ cortex and the effects of electroacupunc-ture (EA) intervention on its expression. Methods 40 healthy adult male Sprague Dawley (SD) rats were equally randomized into Control group,Sham-operation group , Model group and EA group. The model group and EA group with middle cerebral artery occlu-sion were successfully established by an improved Longa procedure.EA was used 90min after anesthesia revival for the EA group.The rats in the EA group accepted the EA treatment for 14 days,once a day. After 14days, immunohistochemistry and Western blot were used to detect expression of the RhoA and ROCK in the brain.Results Compared to the model group,the.neurological.dysfunction.of. EAlgroupkrecoveredjsignificantly(p&lt;0.05).Immunohistochemical staining and western blot both indicated that the expression levels of RhoA and ROCK in EA group were less than those in model group (p﹤0.05 ).Conclusion After focal cerebral infarction RhoA and ROCK expresstionmonmratsncortexnwould;significantly.increased.Giving electroacupuncture interventions can down regulate its ex-pression, this may be one of the mechanisms of EA promoting axonal regeneration after cerebral infarction.
