摘要
目的探讨大鼠脑缺血再灌注损伤后RGMb(repulsive guidance molecule b)表达的变化及RGMb在体外对轴突生长的作用。方法①取出生后7-9d的Wistar大鼠提取内嗅皮层(entorhinal cortex,EC)细胞,分为:正常对照组和RGMb(10μg/ml)处理组,采用细胞培养和免疫荧光染色等方法,检测在体外培养中RGMb处理的新生大鼠EC细胞轴突长度的变化。②取正常8w的Wistar大鼠10只,随机分为:假手术组和脑缺血再灌注48h组,各5只。采用Western blot方法检测脑缺血再灌注48h后海马组织中RGMb表达的变化。结果①在体外培养中RGMb(10μg/ml)能抑制新生大鼠EC细胞轴突的生长。②脑缺血再灌注48h后成年大鼠海马组织中RGMb的表达明显上调。结论脑缺血再灌注损伤后RGMb表达明显增加,提示RGMb可能参与脑缺血再灌注损伤后的神经再生障碍、炎症和细胞凋亡等病理生理过程。
Objective To explore the alteration of RGMb levels after cerebral ischemia-reperfusion injury and its effect on neurite growth.Methods Cell culture and neurite outgrowth assay were used to detect the neurite growth of EC neurons from Wistar rats at postnatal days 7-9 treated with RGMb.Western blot was used to determine the alteration of RGMb levels in hippocampus 48h after cerebral ischemia-reperfusion in Wistar rats.Results Neurite outgrowth was obviously reduced when the EC neurons were treated with RGMb(10μg/ml)in vitro.The expression of RGMb was significantly increased in the hippocampus of adult rats 48h after cerebral ischemia-reperfusion.Conclusion The upregulation of RGMb levels after cerebral ischemia-reperfusion injury suggests that RGMb might be involved in the pathophysiologic processes of the lack of nerve regeneration,inflammation,apoptosis and so on after cerebral ischemia-reperfusion injury.
