摘要
实验用优化煮沸法提取鸡血液基因组DNA,经核酸蛋白定量仪检测,DNA的OD260/OD280达1.77±0.06,证明所提DNA质量较好,用于PCR扩增,可以得到满意的扩增效果。与酚-氯仿法相比,优化煮沸法避免了用氯仿、酚等剧毒试剂,且节约了时间和成本,是一种较好的提取基因组DNA的方法。
The optimized cooking extraction method was used in this study to extract gallus genomic DNA.OD260 /OD280 was up to 1.77±0.06 by the SmartspecTMplus test.The result showed that the quality of DNA extracted by the optimized cooking extraction method was very good,clear and bright fragments were detected through PCR amplification.Compared with the Phenol-chloroform extraction method,The optimized cooking extraction method is free from chloroform,phenol and other toxic reagents,and saves time and costs,hence a good method for extracting genomic DNA.
