龙眼壳多糖含量的测定及其免疫活性研究 被引量:14

摘要 目的从龙眼壳中提取分离龙眼壳多糖,测定龙眼壳多糖在龙眼壳中的含量,同时探索龙眼壳多糖对体外小鼠脾淋巴细胞的免疫活性的影响。方法采用水提醇沉法提取分离龙眼壳多糖,经酶-Sevage法除蛋白和H2O2氧化法除色素后,进一步用DEAE-纤维素柱层析和葡聚糖凝胶Sephadex G-100柱层析对龙眼壳多糖进行纯化,苯酚-硫酸法测定龙眼壳多糖含量;MTT法测定龙眼壳多糖对ConA诱导小鼠脾淋巴细胞的增殖能力;酶联免疫吸附反应(ELISA)分析龙眼壳多糖对ConA诱导小鼠脾淋巴细胞分泌IL-2的诱生作用。结果龙眼壳多糖总糖含量为51.84%,龙眼壳粗多糖得率为3.22%,经过纯化之后的龙眼壳多糖的糖含量为92.01%。龙眼壳多糖不同剂量组对ConA诱导小鼠脾淋巴细胞的增殖能力有增强作用,对ConA诱导小鼠脾淋巴细胞分泌IL-2有明显的促进作用,均与剂量呈正相关关系。结论龙眼壳多糖能增强小鼠脾淋巴细胞的免疫作用。 Objective To extract and purify polysaccharides (LP)from shell of Dimocarpus longan Lour. , measure the polysaccharide contents in shell of Dimocarpus longan Lour. and investigate LP immune function in vitro . Method Crude polysaccharide was isolated by adding EtOH to the hot water extracts of Dimocarpus longan Lour. and was deproteinized by Sevage method and emzymic method,and were decolourised by H2O2. Phenol-sulfuric acid method was employed in the determination of total sugar and polysaccharide contents in shell of Dirnocarpus longan Lour. The crude polysaccharide was further purified by DEAE-cellulose (OH-) ion-exchange and Sephadex G-100 chromatography. ConA-induced splenic lymphocyte proliferation and activity of secretion of IL- 2 in mice were determined respectively by 3-( 4,5-2dimethylthiazal-2yl) 2,5-diphenyltetrazoliumbromide(MTT) assay and enzymelinked immunosorbent assay (ELISA). Results The total content of sugar and the content of polysaccharides LP were 51.84 % and 3.22% ,respectively,and the content of LP were 92.01 % after purification. The ConA-induced spleen lymphocyte proliferation and the ConA-induced secretion of IL-2 in the different doses of LP were decreased and enhanced in dose-dependent manner. Conclusion LP is able to enhance immune function of spleen lymphocytes.
