摘要
目的:应用细胞微囊化方法、病理技术研究微囊化大鼠胰岛细胞与睾丸Sertoli细胞联合移植对糖尿病小鼠血糖的影响及Sertoli细胞对共移植的胰岛细胞的免疫豁免作用。方法:将30只小鼠随机分为2组,每组15只。第一组为胰岛细胞微囊化移植组,第二组为胰岛细胞与睾丸Sertoli细胞联合微囊化移植组,建立糖尿病模型后,两组均采用腹腔注射的方法将微囊植入糖尿病小鼠体内,术后两组均不使用免疫抑制剂,定期检测小鼠的血糖情况,比较两组之间胰岛细胞有功能存活时间的不同以及微囊的病理学改变。结果:胰岛细胞与睾丸Sertoli细胞联合微囊化移植组与胰岛细胞微囊化移植组相比,联合移植组胰岛细胞存活时间长于单纯移植组(P<0.05),囊周淋巴细胞侵润程度轻于单纯胰岛细胞移植组。结论:睾丸Sertoli细胞对共移植的胰岛细胞具有免疫豁免作用,可以延长胰岛细胞有功能存活时间,减轻排斥反应,降低囊周淋巴细胞侵润程度。
Objective: To investigate the immune exemption for islet grafts induced by cotransplanted testicular Sertoli cells in rats and effect on blood glucose by co-microencapsulated rat islet cell and testicular sertoli cell through microcapsulation and pathyologic tecnology. Methods: Thirty mice were randomly divided into 2 groups with 15 in each one. Mice in the first group were transplanted microcapsulated islet and that in the second group were trasplated micro-capsulated islet and testicular sertoli cell. The microcapsules were all transplated into abdominal cavity of diabetic recipient for mice of two groups. Immunosuppressant was not ap- plied and blood glucose was detected after operation so as to compare different survival time and pathologic al- teration of microcapsule. Results: The survival time of islet in 2 groups was different. The time of cotransplantation was longer than that of simple cotransplantation(P〈0. 05)and infiltrated extent for lymphocyte of capsulated periphery in mice of group 2 was more gently than that in group 1. Conclusion:Testicular Sertoli cell can induce immune exemption for eotran-splanted islet; prolong the survival time of islet palliate rejection and degrade infiltration extent of lymphocyte of capsulated periphery.
