摘要
目的:采用HPLC测定野葛和粉葛两种葛根中葛根素的含量。方法:以十八烷基硅烷键合硅胶为填充剂,以甲醇-水(25:75)为流动相,检测波长为250nm。结果:通过方法学考察,取样量在0.16~0.80μg(r=0.9999)范围内呈良好线性关系,加样回收率为97.88%(n=6)RSD为2.4%.结论:方法系统,操作简单,数据准确,重现性好,可作为葛根配方及加工原料选择依据及质量标准的含量测定方法。另外。
Objective: To measure the puerarin in Pueruria lobata (Wild.) Ohwi and Pueraria thonsonii Benth via the HPLC determination. Methods: The puerarin is measured with C18 as a filler, methanol-water (25:75) as a mobile phase and a 250nm-wavelengthed detection. Results: Via the methodological survey, the sample quantity, within the interval 0.16 ~0.80μg (r = 0.9999), has a properly linear relationship with the peak area, and the percent recovery is 97.88 % (n = 6), while RSD is2.4%. Conclusion: In the experiment, the method is systematic, the operation is simple, the data is accurate, and the repeatability is appropriate. Therefore, this experiment, earl be used as the measuring method of the basis of the measure, the raw material selection of the puerarln and the standard of the quality of the Pueruria products. Besides, Pueruria lobata (Wild.) Ohwi contains more puerarin than Pueraria thonsonii Benth. Conclusion: The method the system, the operation simple, accurate data with good repeatability. Content higher, This can be used as Pueraria quality standards and determination methods .
