摘要
目的研究脑得生软胶囊和片剂在家兔体内的药动学及相对生物利用度。方法本试验采用单剂量、双周期试验设计,每只家兔在24h内取血10次,血浆样品用甲醇进行蛋白质沉淀,采用反向高效液相色谱法进行测定。结果葛根素在0.6-9.0μg/mL范围内线形关系良好,回归方程为:Y=0.11708+0.65268X,精密度<15%,准确度在93.3%-102.3%之间,提取回收率>80%,相对生物利用度为(106.7±10)%。结论本方法简单、准确,灵敏,能应用于脑得生片和脑得生软胶囊中葛根素的相对生物利用度研究。
Objective To study the drug kinetics and the relative bioavailability of Naodesheng soft capsule and tablet in rabbit. Methods Single dosage, double period design were utilized in this study. We sampled rabbit blood 10 times within 24 hours and then determined with RP-HPLC after the sample underwent protein precipitate with methanol. Result At range of 0. 6μg/mL-9. 0μg/mL puerarin turned out a good linear dependent relation, with the regression equation： Y=0. 11708＋0. 65268X, precision of analysis was less than 15%, accuracy between 93. 3% and 102. 3%, and the extraction recovery rate over 80%, the relative bioavailability was （106. 7±10） %. Conclusion The method in this study is proved simple, accurate, sensitive for availability of utilization for researches on relative bioavailability of puerarin in Naodesheng tablets and soft capsule.
