摘要
目的:观察首乌、洁脉冲剂及其组分对平滑肌细胞体外药敏效应。方法:从“复方-单味药-活性成分”对比分析入手,采用MTT法测定首乌、洁脉冲剂、大黄素及大黄酸对平滑肌细胞增殖抑制作用。结果:首乌、洁脉冲剂与大黄素的有效剂量范围为其血药浓度的25~125倍;大黄酸有效剂量范围为其血药浓度的12.5~75倍。洁脉冲剂、大黄素对平滑肌细胞增殖抑制率分别为21.54%~96.89%,24.6%~94.58%;而首乌、大黄酸效果较差,抑制率分别为16.56%~86.57%,14.38%~85.64%。并且随作用时间延长、药物浓度的增加抑制率都有一定的提高。结论:本研究为首乌、洁泳冲剂、大黄素及大黄酸的医学应用提供了理论依据,为中药复方药效学研究提供了一种可借鉴的方法。
MTT colormetry is used to decide the effective scope of Radix Polygoni Multiflori, JIEMAI Granules, emodin and rhein to inhibit smooth muscle cells proliferation. Blood drug density is counted according to the dose of row Radix Polygoni Multiflori and JIEMAI Granules. The results show that the effective scope of Radix Polygoni Multiflori, JIEMAI Granules, emodin are from 25times to 125times of its concentration in blood, and rhein is from 12.5times to 75times. The inhibition rate of smooth muscle cells by Radix Polygoni Multiflori is from 16.5% to 86.57%, JIEMAI Granules is from 21.54 to 96.89%, emodin is from 24.6% to 94.58%, and rhein is from 14.38% to 85.64%. JIEMAI Granules and Emodin are more effective than Radix Polygoni Multiflor and rhein in inhibitting smooth muscle cells growth. This research provides theoretical basis of medical application of Radix Polygoni Multiflori, JIEMAI Granules, emodin and rhein.
