大鼠拟“衰老”胸腺细胞功能退化的机制探讨及左归丸的作用 被引量:1

摘要 目的:探讨皮质酮致大鼠胸腺细胞功能退化可能的机制及左归丸的药理作用。方法:大鼠胸腺细胞体外培养,分正常对照组、模型组、左归组。MTT法检测细胞增殖情况,PI染色结合FCM分析周期分布,Real time PCR和Western blotting法检测相关因子表达变化。结果:皮质酮显著抑制细胞增殖,细胞阻滞于G1期,p16表达显著升高(P<0.01),cdk4、IL-2、IL-2Rα蛋白表达降低(P<0.05)。与模型组比较,左归组G1期细胞数减少,p16表达降低,IL-2、IL-2Rα蛋白表达升高(P<0.05)。结论:左归丸通过抑制p16表达,促进细胞增殖,延缓皮质酮致胸腺依赖性免疫功能退化的进程。 Objective :To investigate the accommodation and the molecular mechanism of Zuogui Pill (ZGP) on the proliferation and immuno function of rat tbymocytes (MSTM) in vitro. Method: Thymocytes were divided into three groups : (A) control group ; (B) model group ; ( C ) ZGP group. The effect ZGP with corticosterone on the proliferation of thymocytes was detected by MTT. Cell cycle was measured by PI staining and FCM. The expression of p16, cdk4, IL-2, IL-2R were measured by Real-time PCR and Western Blotting. Resuhs:Corticosterone had some inhibitory effect on thymocytes,arrested cell cycle at G1 phase, up-regulated the expression of p16, down-regulated the expression of IL-2, IL-2R. Comparing with model group, ZGP group showed higher expression of IL-2 and IL-2R and lower expression of p16 (P 〈 0. 05). Conclusions : The possible mechanism of immuno-regulation effect of ZGP is to regulate the expression of p16, IL-2, IL-2Rα in MSTM, postpone the thymus dependented immune functional deterioration induced by corticosterone.
