摘要
目的:探讨人各级别胶质瘤中β-catenin的表达及其与胶质瘤发生发展的关系。方法:采用RT-PCR、WesternBlot及免疫组化方法,检测正常脑组织和各级别胶质瘤标本中β-catenin的表达。结果:随胶质瘤级别的升高,β-cate-nin在mRNA和蛋白水平的表达均逐渐升高,与正常对照存在统计学差异(P<0.05)。在高级别胶质瘤中的β-catenin的表达明显高于低级胶质瘤(P<0.01)。免疫组化与上述结果类似,各级别胶质瘤同正常对照有统计学差异(P<0.05)。结论:β-catenin表达水平随胶质瘤恶性程度的增加而增加,提示β-catenin可能参与了胶质瘤的发生发展过程。
Objective:To investigate the expression of β-catenin in human glioma and explore the relationship between the expressions of β-catenin and tumorgenesis as well as progressions of glioma.Methods:RT-PCR,western blot and immunohistochemistry were empolyed to detect the expression of β-catenin in glioma patients and normal controls.Results:With the increased grade of glioma,the mRNA and protein expression level of β-catenin was gradually increased,which had a significant difference compared to normal controls(P〈0.05).Meanwhile,the expression of β-catenin in high-grade gliomas was significantly higher than that in the low-grade ones(P〈0.05).Moreover,immunohistochemical results were similar to the above.There was a significant difference in β-catenin expression level between glioma patients and the controls(P〈0.05).Conclusion:The expression level of β-catenin is correlated with the stage of glioma,suggesting that β-catenin might be probably involved in the gliomagenesis and development of glioma.
