摘要
几乎所有的人类肿瘤都存在多基因损伤,药物治疗很难只作用于癌基因本身,而不影响相对应的未发生突变的原癌基因及其相关因子。最近发现的RNA干扰和小干扰RNA(small interfering RNA,siR-NA)在哺乳动物细胞中的应用,为肿瘤特异性治疗途径提供了新的工具。因其具有高度特异性和高效性,siRNA可能成为基因治疗的新型抗肿瘤药。总结siRNA在肿瘤中的研究成果,包括在细胞培养和动物模型对癌基因的抑制作用、有效的siRNA传递系统的开发及其治疗潜能发展的研究。
Almost all human cancers have accumulated multiple genetic lesions. It is very difficult to target an essential oncogene with drugs without affecting the corresponding nonmutated protooncogene or related factors. The recent discovery of RNA interference and the application of small interfering RNAs in mammalian cells provide new tools for more cancer-specific therapeutic approaches. Because of their highly specificity and efficiency, siRNAs might be a new class of anti-gene medicines for gene therapy applications. This review summarizes the results obtained with siRNA in cancer, including functional validation of tumorigenic genes in cell culture and animal tumor rnodels,effective siRNA delivery systems and the potential for therapeutic development.
