不同时间窗吻合大鼠坐骨神经后应用CNTF对脊髓运动神经元内Ca^(2+)的作用 被引量:2

Effect of CNTF on Ca^(2+) level in spinal cord motor neurons after sciatic nerve anastomosis at different time points post-injury

摘要 目的:研究睫状神经营养因子(CNTF)对坐骨神经损伤延期手术后对相应脊髓Ca2+作用和脊髓神经损伤的再生修复作用.方法:大鼠双侧坐骨神经切断后延期即刻、3,7,14,21d后行神经外膜端-端吻合术,给予CNTF治疗.吻合术后饲养3d后处死检测脊髓Ca2+浓度,饲养28d后处死行Tunel染色凋亡细胞计数、HE染色观察L4~6脊髓神经元形态、神经吻合口部位神经纤维镀银染色形态学观.结果:用药组TUNEL阳性细胞数目比未用药组、盐水组少,用药组在3个大组中凋亡数目少且均与其他3组有统计学差异(P<0.05),用药组7d凋亡数目较盐水组、非用药组低;Ca2+质量浓度较未用药组、盐水组增幅减低;用药组即刻、3d、14d、21d亚组28d处死时可见部分神经纤维通过吻口,7d通过吻合口更多,但排列不整齐,而未治疗组和对照组的神经纤维通过吻口数量较治疗组不明显.结论:坐骨神经在不同时间切断吻合后均可诱导脊髓前角运动运动神经元的凋亡;大鼠坐骨神经离断吻合后,不同时间窗吻合术后应用CNTF可以降低急性期脊髓组织Ca2+离子水平,可能起到减少脊髓运动神经元凋亡的发生、保证周围神经轴索再生数量的作用. AIM:To research on motor neurons protection of spinal cord segment corresponding to traumatic sciatic nerve injury by ciliary neurotrophic factor(CNTF).METHODS:The sciatic nerves of both sides at rats were transected and repaired after a delay of immediately,3,7,14,21 d and CNTF treatment was given.They were sacrificed after 3 d and detected spinal Ca2+ concentration.Sacrificed them after 28 d and counted apoptotic cells with staining,HE detected morphology changes of L4-6 spinal neurons,morphology observation of nerve fibers on nerve anastomosis.RESULTS:The number of TUNEL positive cells in five groups of medication group was less than that in non-medication group and saline group.The number of apoptotic cells in 7 d group was smaller than that in non-medication group and saline group and was the smallest compared with the other four groups(P0.05).While increase amplitude of Ca2+ ionic concentration and plasma endothelin level was lower.some nerve fibers passed the cut in medication group on 28th day.More fibers were found in 7,14 d group,but irregularly.Fewer fibers were found in non-medication group and saline group.CONCLUSION:That sciatic nerve is transected and repaired at different time can cause apoptosis of spinal cord anterior horn motor neurons.After the sciatic nerves at rats were transected and repaired,early use of CNTF can decrease Ca2+ in acute spinal tissues and keep ion balance in and out of cells.This may decrease apoptosis of spinal motor neurons and keep regeneration of peripheral nerve axon.
