摘要
本文探讨用分光光度法测定血红素中铁离子的含量。通过对血红素进行硝化处理,将其中铁离子游离出,并与适量的邻菲罗啉在pH为4.6的缓冲液中定量生成橙红色络合物,用分光光度法在510±1nm处测定其含量。结果显示,铁离子与邻菲罗林生成的橙红色络合物在510±1nm波长处有最大吸收,且在0.4~2.8μg/ml范围内,浓度与吸收度符合Beer定律,相关系数r=0.9999,方法平均回收率X=98.87%,RSD=0.58%。本法结果准确,方法可行,适用于医院、药厂生产的各种血红素制剂的质量分析与检验,并可为医生用药提供依据。
Iron-ion in Hematin was determination by spectrophotometry at wavelengths of 510± 1nm.The result showed a good linear relationship in the concentration range of 0. 4~ 2, 8 μg/ml (r =0. 9998).The average recovery rate was 98. 87 %,the coefficient of variation was 0. 58 %. The method is feasible,the result is accurate. It is suitable for analysis of hospital preparation and pharmaceutical factory.
