摘要
采用超声波法辅助提取、HPLC测定的方法,研究了豚草不同部位绿原酸的含量.并与烟草、金银花和几种南京郊区常见杂草的绿原酸含量进行了比较,将提取剩余物用间型脉孢菌进行了生物转化.实验结果表明,豚草各部位的绿原酸含量以叶最多,为1.160 mg/g;花和茎次之,分别为0.559和0.494 mg/g;豚草叶绿原酸以7月份含量最高.在50℃时用超声波辅助提取,作用时间以20 m in为宜.用间型脉孢菌转化提取剩余物,物料经生物转化后,富含纤维素酶、蛋白酶和类胡萝卜素,其中纤维素酶中CMC酶活性为5 503.9 U/g,蛋白酶活性为7 926.4 U/g,类胡萝卜素含量为126.6μg/g.提取酶和类胡萝卜素后的物料可作为生产有机肥的原料,使物料得到全部利用.
The content of chlorogenic acid in different parts of Ambrosia artemisiifolia L. was studied by HPLC analysis method and ultrasonic-assisted extraction. Chlorogenic acid contents of Ambrosia artemisiifolia L. , Nicotiarta tabacum, Lonicera japonica Thunb. and several common weed of Nanjing were compared. The extraction residues were treated by Neurospora intermedia. The results showed that the contents of chlorogenic acid in different parts of Ambrosia artemisiifolia L. were different. Leaves were 1. 160 mg/g, flowers and stems were 0. 559 mg/g and 0. 494 mg/g respectively. The ehlorogenic acid content of Ambrosia artemisiifolia L leaf is the highest in July. When extracted by ultrasound-assisted method at 50℃, the proper extraction time was 20 min. After bioconversion by Neurospora intermedia, the extraction residues contain carotenoid and abundance of enzymes such as cellulase and protease, the yields of carotenoid, as high as 126.6 μg/g, enzyme activity of CMCase and Protease were 5 503.9 U/g and 7 926. 4 U/g, respectively. After the enzyme and carotenoid were extracted from fermentation products, the residue was entirely utilized as organism fertilizer totally.
