摘要
通过受精卵原核显微注射的途径,产生带有可诱导型丙肝病毒非结构蛋白3基因(nonstructural3gene)的重组载体的转基因小鼠。采用PCR和基因组Southernblot杂交方法,在21只新生鼠中筛选到3只建立者小鼠。其中2只与非转基因小鼠进行交配,分别得到8只和6只F1代小鼠。进而对其基因组DNA进行检测,发现其中4只为转基因阳性个体。这些小鼠的获得,为ns3转基因品系的建立和NS3蛋白的生物学作用研究准备了条件。
To generate transgenic mice, the recombinant plasmids inserted the ns3 gene of hepatitis C virus were introduced into the male pronuclei of mouse fertilizer eggs by microinjection. By using polymerase chain reaction(PCR) and Southernblot hybridization method we obtained three mice(Founders) and four mice(F1) harbouring the ns3 gene among 21 offsprings born by the pseudopregnant mice and 14 individuals generated from the Founders mated with other nontransgenic mice, respectively. These transgenic mice would be helpful for the development of ns3 transgenic mice line and the study for the biological function of NS3 protein in vivo.
