摘要
目的:人中性粒细胞防御素具有广谱杀菌活性。将人中性粒细胞肽(HNP)-1基因与J链基因融合转染其他细胞有可能使HNP表达而发挥杀菌作用。文中分析HNP1与J链融合基因在COS-7细胞中表达蛋白的影响因素。方法:应用免疫印迹技术,注意控制影响蛋白转印的关键条件,分析影响J-HNP1蛋白表达的技术因素。结果:成功检测到融合基因J-HNP1在细胞内的表达。结论:采用Western blot法可检测到J-HNP1在细胞内的表达并可分泌到细胞外。
Objective:Defensins from human neutrophils exhibit a broad antimicrobial activity. The transfection of the gene fused by human neutrophil peptide 1 (HNP1) with the J chain into other cells may induce the in vitro expression of HNPI to play antimicrobial roles. The authors analyzed the influencing factors on the expressed proteins of HNP1 and the J chain gene in COS-7 cells. Methods : Western blot was used to analyze the technical factors influencing the HNP1 protein expression. Results: Recombinant J-HNP1 was expressed in the cells successfully. Conclusion: The expression of J- HNP1 can be detected inside and outside the cells by Western blot.
