应用激光捕获显微切割及蛋白质组学技术筛选鼻咽癌间质相关蛋白

Screening for the Stroma-related Proteins in Nasopharyngeal Carcinoma by Laser Capture Microdissection and Proteomic Analysis

摘要 目的筛选与鼻咽癌(NPC)发生发展相关的特异性间质蛋白。方法采用激光捕获显微切割技术(LCM),获取高纯度的NPC间质和正常鼻咽黏膜(NNM)间质,二维凝胶电泳(2-DE)及质谱技术,分离鉴定间质相关蛋白。W estern b lot及组织芯片免疫组织化学技术,验证差异蛋白S100A9在鼻咽癌间质和正常鼻咽黏膜组织中的表达水平。结果建立了鼻咽癌间质和正常鼻咽间质的2-DE图谱,高通量筛选与肿瘤发生相关的间质蛋白,质谱鉴定得到60个差异蛋白。W estern b lot及免疫组织化学技术验证结果与蛋白质组学结果一致。结论这些差异表达的蛋白将有助于揭示鼻咽癌细胞和周围间质的关系。对间质蛋白功能的进一步研究,将有助于解析间质在肿瘤发生发展中的作用机制,从间质途径寻找肿瘤治疗的靶标。 Objective To screen the stromal- related proteins involved in nasopharyngeal carcinoma (NPC) carcinogenesis. Methods Laser capture microdissection (LCM) was performed to purify stromal cells from the NPC and norreal Nasopharyngeal mucosa (NNM) , respectively. The protein expressed profiles of the stroma of NPC and NNM were compared using two - dimensional electrophoresis and the differential proteins between the 2 types of stroma were identified by MALDI - TOF - MS. Western blot and immunohistochemistry of tissue microarray were used to confirm the expression of the differential protein S100A9 in NPC and NNM. Results 2 - DE patterns of stroma of NPC and NNM were established,and 60 differential proteins in the NPC and NNM were identified. The differentially expressed level of S100A9 was confirmed by Western blot and immunohistochemistry. Conclusion These differentially expressed proteins in the stroma of NPC and NNM may be useful for understanding the relationship between NPC cells and their surrounding microenvironmerit. Further studying of these proteins will be helpful to elucidate the mechanisms of NPC carcinogenesis and find new targets on therapy of NPC.
