摘要
[目的]寻求杂交朱顶红鳞茎不定芽诱导的最佳配方,以提高杂交朱顶红组培扩繁系数。[方法]以杂交朱顶红鳞茎为外植体,探讨不定芽诱导的最佳部位、最佳激素浓度,并比较不同品种的杂交朱顶红对不定芽产生的影响。[结果]朱顶红鳞茎诱导不定芽最适部位为鳞茎下部,苹果诱导频率为87.12%,杂交后代18号诱导频率为31.55%;不定芽发生最适培养基为MS+TDZ0.5mg/L+NAA0.1mg/L,无论是诱导频率还是每外植体诱导不定芽数均为最高,苹果诱导频率为100.0%,杂交后代18号诱导频率为70.1%;不同品种不定芽分化差异较大,杂交后代比国外引进品种更难诱导不定芽。[结论]为朱顶红的种球国产化提供了强有力的技术支持。
[ Objective ] The research aimed to find optimum formula for adventitious bud induction of Hippeastrum hybridum bulb and improve its tissue culture propagation coefficient. [ Method i With H. hybridum bulb as explant, the best part, hormone concentration of induction were discussed,and the effects of different species H. hybridum on adventitious were compared. [ Result I The optimum part was the lower of bulb. The frequency of apple induction was 87.12% ,and filial generation No. 18 was 31.55%. The optimum culture medium was MS + TDZ 0.5 mg/L + NAA 0. 1 mg/L, in which no matter induction frequency or number both are the highest. The frequency of apple induction was 100.0% ,and filial generation No. 18 was 70.1%. Different species have significant differences. Filial generation was harder than imported varieties in adventitious bud induction. [ Conclusion ] The research provided a powerful technical support for H. hybridum bulb producing in our country.
