肝细胞癌中RhoC mRNA的表达及其临床意义

EXPRESSION OF RHOC GENE IN HEPATOCELLULAR CARCINOMA AND ITS CLINICAL SIGNIFICANCE

摘要 目的:探讨RhoC mRNA在人肝细胞癌(Hepatocellular carcinoma,HCC)组织中的表达及其与HCC复发转移的关系。方法:用荧光定量PCR技术检测82例HCC及其癌旁肝组织,16例正常肝组织中RhoC mRNA的表达情况。分析mRNA表达量与临床资料之间的关系。结果:RhoC mRNA在HCC、癌旁组织和正常组织中均可检测到表达,表达量分别为1.18±0.77、0.67±0.29和0.38±0.18,组间表达量比较差异有统计学意义(P<0.05),RhoC mRNA在HCC组织中的表达水平明显高于癌旁肝组织中的表达水平,两者差异有统计学意义(P<0.05);RhoC mRNA在HCC中的表达量与结节数目多,门静脉癌栓,术后复发转移相关(P<0.05),RhoC mRNA高表达的患者术后5年生存率明显低于RhoCmRNA低表达的患者(P<0.05)。结论:RhoC mRNA的过表达可能与HCC的发生及复发转移有关,有可能做为HCC患者术后复发转移的预测指标。 Objective:It has been suggested that one member of the Rho gene family,RhoC,is related to the progression of human hepatocelular carcinoma(HCC).This study is to elucidate the correlation of Rho overexpression with invasion and metastasis of HCC.Methods:Real-time reverse transcriptase-polymerase chain reaction was performed to observe RhoC expression in 82 cases of HCC tissues,adjacent non-cancerous liver tissue and 16 normal liver tissue.Result:The expression of RhoC mRNA was found in all HCC,adjacent non-cancerous liver and normal liver tissues.The expression level of RhoC mRNA was significantly higher in tumor tissue than in adjacent non-cancerous liver tissue and normal liver tissue(1.18±0.77 vs 0.67±0.29 vs 0.38±0.18,P<0.05).Overexpression of the RhoC gene was significantly correlated with such clinic-pathological findings as portal vein invasion,number of primary tumor nodules and metastatic lesions(P<0.05).The survival rate of patients whom with high expression of RhoC mRNA was obviously lower than that of the low expression patients(P<0.05).Conclusion:Our investigation revealed that RhoC positively correlated to the prognosis of HCC patients.The expression of RhoC in HCC tissues could be regarded as a valuable indicator in estimating the prognosis of HCC patients.
