不同来源白油佐剂质量分析及其制备的禽流感疫苗安全性与免疫效力研究 被引量:24

Quality Analysis of Oil Adjuvants from Different Origins and Safety and Immune Efficacy Comparison of Inactivated Avian Influenza Vaccines Prepared with These Oil Adjuvants

摘要 通过抽取3种来源注射用白油LH、MC、HZ,按照《中华人民共和国兽药典》附录"注射用白油质量标准"及企业内控质量标准要求进行成分分析,3种白油在黏度、稠环芳烃含量等指标方面存在差异;将上述来源白油佐剂制备的共计6批禽流感灭活疫苗分别以0.3mL/只及0.6mL/只剂量,经胸部肌肉和颈部皮下注射途径接种SPF鸡,接种后8、13、20、31、41、48d分别进行疫苗的安全性检查和免疫效力检测。疫苗安全性检验结果表明:疫苗免疫后均导致注射部位出现局部病变和不吸收现象,恢复较快的是黏度为5.7mm2/s的LH1、LH2组及黏度为11.2mm2/s的HZ组,黏度为7.26mm2/s、稠环芳烃含量偏高的MC组吸收稍差;疫苗效力检验结果表明:黏度较低的LH1~LH4组疫苗免疫后抗体产生时间、维持水平均优于MC组、HZ组。 Systemic detection of white oil adjuvants from three origins(named LH,MC,HZ)were carried out according to China Veterinary Pharmacopoeia (2005) and the internal control criterion of QYH. There were differences in viscosity and polycyclic aromatic hydrocarbon condensed aromatics content. 6 batches of the inactivated avian influenza vaccines prepared from these oil adjuvants were inoculated by subcutaneous route in the base of neck or chest muscular route of SPF chickens,each with 0.3 mL or 0.6 mL inoculum. After 8,13,20,31,41 and 48 days,the vaccines were tested in safety and potency. The safety test result showed that three experimental groups all appeared local pathological change and non- absorbed in the inoculation parts. LH1 and LH2 groups with the viscosity of 5.7 mm2/s and the HZ group with the viscosity of 7.26 mm2/s recoveried quickly,the less absorption was MC group with higher content of polycyclic aromatic hydrocarbon. The potency test result indicated that HI antibody appeared earlier and remained a longer time for LH1 to LH4 group with a lower content of viscosity than those for MC and HZ groups.
