摘要
目的:观察胰腺组织及其癌变标本中基质金属蛋白酶(MMPs)及金属蛋白酶组织抑制因子(TIMPs)基因的表达,以探讨MMPs及TIMPs在肿瘤侵袭和转移过程中的作用。方法:采用cDNA探针(MMP2、MMP9、TIMP1)对两株胰腺癌细胞(PaTu8988、PaTu8902)、15例胰腺癌手术切除标本和8例正常胰腺组织标本进行Northern杂交。结果:胰腺癌组织和PaTu胰腺癌细胞MMP2、MMP9和TIMP1的基因表达水平明显高于正常胰腺组织,其趋势为:MMP2≈MMP9<TIMP1。结论:在胰腺癌组织中,MMP2、MMP9、TIMP1可能参与了组织成分较强的降解反应过程,从而使肿瘤更易侵袭和转移;肿瘤和间质细胞似乎都能产生MMPs和TIMPs。
Objective: To evaluate the expression of matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMPs) in human pancreatic tissue and pancreatic cancer specimens in order to explore the role of MMPs and TIMPs in the neoplastic transformation and invasiveness. Methods: Two pancreatic cancer cell lines (PaTu8988, PaTu8902), 15 cases of surgical pancreatic cancer specimens and 8 normal pancreatic tissue specimens were tested by Northern blot with cDNA probes (MMP2, MMP9 and TIMP1). Results: The expression of MMP2, MMP9 and TIMP1 genes in pancreatic cancer tissue and PaTu pancreatic cancer cell line was significantly higher than that in normal pancreatic tissue and its trend was MMP2≈MMP9<TIMP1. Conclusion: The results suggest the potent role of MMPs and TIMPs in the processes which lead to metastasis and local invasiveness of human pancreatic cancer cells.
