摘要 目的:分析果蝇zeste基因增强子同源物2(enhancer of zeste homolog 2,EZH2)和血管内皮生长因子(vascular endothelial growth factor,VEGF)在原发性肝细胞性肝癌(primary human hepatocellular carcinoma,HCC)中的表达及其临床意义。方法:应用免疫组化S-P法检测HCC 50例、肝硬化30例、正常肝组织10例中EZH2、VEGF蛋白的表达。结果:HCC、肝硬化和正常肝组织的VEGF蛋白阳性表达率分别是24.00%(12/50)、66.67%(20/30)、10.00%(1/10),三者之间的差异有统计学意义(x^2=10.34,P<0.05),且VEGF蛋白在肝硬化中的表达明显高于在HCC的表达(x^2=11.22,P<0.05);HCC、肝硬化和正常肝组织的EZH2蛋白阳性表达率分别是88.00%(44/50)、46.67%(14/30)、30.00%(3/10),三者之间的差异有统计学意义(x^2=20.63,P<0.05),且EZH2蛋白在HCC中的表达明显高于在肝硬化和正常肝组织中的表达(x^2=16.07,x^2=14.07,P<0.05)。结论:EZH2、VEGF在HCC中的表达关系不密切(P>0.05);但EZH2和VEGF异常表达与HCC的发展密切相关,在HCC的发展过程中起非常重要的作用。 Objective To explore the expressions of EZH2 and VEGF in hepatocellular carcinoma (HCC) and their clinical significance. Methods The expressions of EZH2 and VEGF were detected by immunohistoehemistry in 50 patients with HCC, 30 cirrhotic patients, and 10 specimens of normal liver tissue. Results The expression rate of VEGF protein differed significantly among the three different tissues (24.00% in HCC, 66.67% in cirrhosis, and 10.00% in the normal tissue; Х^2 = 28.44; P 〈 0.05). VEGF expression was markedly higher in the cirrhotic tissue than in the cancer tissue (Х^2 = 11.22, P 〈 0.05). The expression rate of EZH2 protein also differed significantly among the three tissues (88.00% in HCC, 46.67% in cirrhosis, and 30.00% in the normal tissue; Х^2= 20.63; P 〈 0.05). EZH2 expression was notably higher in the cancer tissue than in the cirrhotic and the normal tissue (Х^2 = 16.07 and Х^2 = 14.07, respectively; P 〈 0.05). Conclusions The expression of EZH2 is not closely associated with that of VEGF in hcpatoeellular carcinoma, whereas the abnormal expressions of EZH2 and VEGF are closely associated with the development of the malignancy.
