摘要
目的探讨斑蝥酸钠维生素B6与人肝癌细胞HepG2凋亡的关系。方法采用MTT法动态观察不同浓度的斑蝥酸钠维生素B6对HepG2细胞生长的抑制率,采用流式细胞术分析不同浓度斑蝥酸钠维生素B6作用下HepG2细胞的凋亡率,荧光显微镜观察HepG2细胞凋亡的形态学改变。结果斑蝥酸钠维生素B6可明显抑制HepG2细胞生长及诱导细胞凋亡(P<0.05),且基本随着其浓度的增加以及作用时间的延长,抑制率及凋亡率逐渐升高,差异有统计学意义(P<0.05),提示斑蝥酸钠维生素B6对HepG2细胞生长的抑制效应及诱导凋亡作用具有浓度依赖性和时间依存性。荧光显微镜下可见经斑蝥酸钠维生素B6处理后的HepG2细胞具有明显的凋亡形态学改变。结论斑蝥酸钠维生素B6可能通过诱导凋亡的途径抑制人肝癌细胞HepG2的生长。
Objective To study the growth inhibision effect of disodium cantharidinate and Vitamin B6 injection on hepatocellular carcinoma cell line HepG2. Methods Different concentrations of disodium cantharidinate and Vitamin B6 injection were applied on the hepatocellular carcinoma cell line HepG2. The inhibition rate of HepG2 growth was measured by MTT assay. The apoptotic rate was detected with flow cytometry, and the morphology of apoptosis was observed by fluorescence microscope. Results The growth of HepG2 cells was significantly inhibited as well as the apoptosis of HepG2 cells was significantly encouraged by disodium cantharidinate and Vitamin B6 injection (P〈0. 05), and as its concentration was increased and the applied time was prolonged, the rates of inhibition and apoptosis increased gradually, which both had statistically differences (P〈0.05). It suggested that disodium eantharidinate and Vitamin B6 injection effected on inhibition of growth and induction of apoptosis of HepG2 cells in a concentration dependent and a time-dependent. HepG2 cells with obvious morphological changes of apoptosis induced by disodium cantharidinate and Vitamin B6 treatment could be seen under the fluorescence microscope. Conclusion Disodium cantharidinate and Vitamin B6 injection may inhibit the growth of HepG2 cells through inducing apoptosis of HepG2 cells.
