摘要
目的探讨TPX2在食管鳞癌及其癌前病变中的表达和意义。方法采用SP免疫组织化学方法检测TPX2和Aurora A蛋白在62例食管鳞癌、31例不典型增生组织及62例正常食管黏膜中的表达水平。结果免疫组织化学结果显示,TPX2蛋白在正常黏膜、癌旁不典型增生和癌组织的表达率依次增高,分别为4.8%(3/62),51.6%(16/31)和85.5%(53/62),三组间比较差异有统计学意义(P<0.05)。TPX2的蛋白表达水平与食管癌的淋巴结转移和浸润深度密切相关,组间比较差异有统计学意义(P<0.01)。Aurora A蛋白在正常黏膜、癌旁不典型增生和癌组织的表达率依次增高,分别为3.2%(3/62),45.2%(14/31)和71.0%(44/62),三组间比较差异有统计学意义(P<0.01),且表达水平与食管癌的组织学分期、浸润和转移相关。结论TPX2、Aurora A基因蛋白的表达可能是食管鳞癌淋巴结转移的危险因素,两者的联合检测可望成为食管鳞癌的早期诊断指标,是判断预后的指标之一。
Objective To study the expression of TPX2 and Aurora A and their significance in squamous cell carcinoma of the esophageal (SCCE).Methods Immunohistochemical staining was used to detect the
expressions of TPX2 in 62 cases of esophageal squamous cell carcinoma specimens,31 cases of adjacent atypical hyperplasia epithelium and 62
cases of normal esophageal epithelium.Reverse transcriptionpolymerase chain reaction analysis for TPX2 and Aurora A was also performed using
tumor tissues from 30 patients with the normal mucos epithelium of esophagus,lesion precancerous and esophageal arcinoma.ResultsImmunohistochemical study showed that the expression rates of TPX2 protein increased by normal esophageal epithelium specimens adjacent atypical hyperplasia epithelium and turns in carcinoma,which were 4.8%(3/62), 51.6 %(16/31)and 85.5%(53/62)respectively.There was a significant statistic difference among the
three groups(P〈0.05).The expression of TPX2 protein was very correlated with the histological grade,infiltrative depth and lymphatic metastasis in esophageal squamous cell carcinoma (P〈0.01).The expression of TPX2 also correlated with lymph node and infiltration.And there was a significant difference between the experimental groups and
