摘要
研究何首乌中二苯乙烯苷成分较佳的分离纯化工艺条件。采用层析柱分离法,选用聚酰胺为吸附剂,食用乙醇为洗脱剂,通过单因素试验和多因素正交试验,研究理想的分离工艺。二苯乙烯苷含量检测采用分光光度法。结果显示:二苯乙烯苷理想分离工艺条件为:食用乙醇体积分数为70%,食用乙醇洗脱流速为1.0 mL/min,洗脱柱高为70 cm,静置吸附时间为20 min。食用乙醇体积分数显著影响二苯乙烯苷的分离效果。聚酰胺柱层析可以较好地分离纯化何首乌中二苯乙烯苷成分。
The ideal separation and purification method of stilbene glucoside from polygonum multiflorum thunb were investigated. Polyamide was used as absorbent, edible ethyl alcohol as eluent, the better separation conditions were researched with single factor experiment and the orthogonal test by column chromatography. The determination of stilbene glucoside was measured by spectrophotometry. Results: The concentration of edible ethyl alcohol was 70%, the flow rate 1.0 mL/min, the column length 70 cm, the absorption time 20 min. The concentration of edible ethyl alcohol was a significant factor. Conclusion: It was feasible to separate and purify stilbene glucoside from polygonum multiflorum thunb.
