摘要
对紫外分光光度法测定柚皮中的黄酮含量及活性测定进行了研究,实验结果表明:紫外检测的最佳波长为360 nm,测定上限可达到8.0 mg/L,下限为0.1 mg/L,回收率为97%,RSD为2%~3%;柚皮提取物具有较强的降低MDA和增加SOD活力的作用,且效果明显;柚皮提取物对脂肪有很强的抗氧化作用,但耐热性不强;80%乙醇洗脱组分A41、B41在0.9g/L时清除DPPH和超氧离子自由基能力与槲皮素相当,但柚皮提取物混合相在低浓度时清除羟基自由基活性最为明显。
The content and activity of flavonoids in pomelo peel were determined using UV spectrophotometry in the study. The results showed that the optimal ultraviolet detection wavelength was 360 nm. The maximum and minimum detection limints were 8.0 mg/L and 0.1 mg/L, respectively. The recovery yield and RSD under the optimal conditions were 97% and 2%-3%, respectively. Extraction ofpomelo peel could remarkably decrease the value of MDA and increase the activity of SOD. The extract ofpomelo peel had a strong antioxidative effect but was heat-liable. The extraction of pomelo peel (0.9 g/L) after diluted by 80% ethanol was found to have the highest clearing activity of DPPH and O2-, which was equal to that of quercetin. The extraction ofpomelo peel mixture at low concentrations had a high activity of clearing the ·OH.
