摘要
[目的]通过对天津市推广种植水稻品种进行组织培养,探索适合天津市推广水稻品种的培养条件,为天津市水稻的遗传改良工作奠定基础。[方法]对5个天津市推广种植的水稻的成熟胚进行离体培养,研究诱导和分化培养基、激素配比、不同接种方式及分化前不同预处理等对其再生的影响。[结果]在NB培养基中添加2.00 mg/L2,4-D和0.50 mg/L6-BA比只添加2.00 mg/L2,4-D的愈伤诱导率高3.79~14.64个百分点;对于津原系列的品种其半粒接种方式愈伤诱导率要高于整粒接种方式;NB+1.00 mg/L6-BA+0.25mg/LNAA+2.00 mg/LKT的分化培养基得到了较高的分化率;干燥处理和预分化均能提高愈伤分化率。[结论]该研究探索影响水稻成熟胚愈伤组织诱导和分化的因素,建立其植株再生体系,为进一步的遗传转化奠定了基础。
[ Objective ] The seeds of rice varieties of Tianjin municipality were conducted for tissue culture to explore the culture condition for rice cultivars of Tianjin municipality and lay the base for genetic improvement of Tianjin municipality. [ Method] The seeds of 5 flee varieties were used to induce the calli in the different medium supplement with different concentrations and combinations of phytohormones and different inoculation methods and different pretreatment before regeneration. [ Result ] The results showed that the callus induction rate in NB medium sup- plemented with 2.00 mg/L 2,4-D and 0.50 mg/L 6-BA was higher than that with 2.00 mg/L 2,4-D. The highest plant regeneration rate could be obtained on the differentiation medium of NB + 1.00 mg/L 6-BA + 0.25 mg/L NAA + 2.00 mg/L KT. Dehydration treatment and pre-differentiation can enhance plant regeneration ability of the calli. [ Conclusion ] This research explored the factors influencing plant regeneration of rice mature embryos and established a set of suitable plant regeneration system, which laid base for flee genetic transformation.
