15-LOX-1和PPARγ间的相互作用及其与肿瘤的关系

摘要 15-脂加氧酶1(15-LOX-1)表达于人的多种组织细胞中,是催化不饱和脂肪酸代谢的关键酶之一,可以将亚油酸、花生四烯酸分别代谢为13-羟基十八碳二烯酸和15-羟基二十碳四烷酸,这些不饱和脂肪酸及其代谢衍生物是过氧化物酶增殖激活受体γ(PPARγ)的天然配体。研究发现,PPARγ以非活性状态在多种肿瘤组织中高表达,其与配体结合活化后,可诱导肿瘤细胞凋亡或者分化,发挥潜在的抗肿瘤作用。以15-LOX-1和PPARγ为靶向的肿瘤防治及其临床应用具有广阔前景。 15-lipoxygenase-1 (15-LOX-1 )is expressed in varieties of human tissues. As one enzyme of the lipoxygenase family, 15-LOX-1 can metabolize linoleic acid and arachidonic acid to13-S-hydroxyoctadienoic acid and 15-hydroxyeicosatetraenoic acid respectively, which are natural ligands of PPARγ. The recent evidences indicate that PPARγ is highly expressed in some tumors with non-active state, which can suppress cell proliferation and induce apoptosis after combining with its ligands. PPARγ and its ligands could be valuable tools in the antitumors. By 15-LOX-1 and PPARγ-targeted research, it is possible to provide means of early detection and diagnosis of cancers and novel therapeutic options for treatment or prevention of tumors in the future.
