摘要
目的观察维奥欣对实验性动脉粥样硬化(AS)大鼠颈动脉壁肿瘤坏死因子α(TNF-α)的影响,探讨其在AS中是否存在抗炎作用。方法60只雄性SD大鼠随机分为假手术组、模型组、阿司匹林组、维奥欣小剂量组和维奥欣大剂量组。大鼠于饲养第7日经左颈总动脉主动脉内膜球囊损伤术。3个月后处死动物,留取标本,测定血脂、行颈总动脉病理形态学分析,用免疫组化及Westen-blot法检测颈总动脉TNF-α。结果与模型组比较,维奥欣大剂量组血清TG、TC、LDL-C水平显著下降,且明显低于阿司匹林组。维奥欣小剂量组与模型组无明显差异;HDL-C水平在治疗组与模型组间无明显差异。颈总动脉健侧各组W/L无明显差异;模型组损伤侧颈动脉内膜明显增厚,局部有斑块形成,W/L明显增大。维奥欣大剂量、小剂量组W/L明显小于模型组,维奥欣大剂量组低于阿司匹林组,维奥欣小剂量组与阿司匹林组间无显著性差异。免疫组化染色及Westernblot检测结果显示,与模型组比较,维奥欣大剂量、小剂量组TNF-α表达水平显著下降,维奥欣大剂量组明显低于阿司匹林组,而维奥欣小剂量组与阿司匹林组间无显著性差异。结论维奥欣可通过减少实验性AS大鼠TNF-α水平,抑制炎症反应而达到抗AS作用。
Objective: To observe the effect of dioscornin on expression of tumor necrosis factor - α(TNF - α) of experiment atherosclerosis(AS)rats and investigate whether its anti - inflammatory action on AS or not. Methods: 60 male SD rats were randomly divided into five groups: sham operation group, model group, aspirin group, dioscornin low and high dosage groups with 12 in each group. After 7d, the balloon - injured carotid artery angioplasty was performed in the rats of all groups except sham operation group. 3 months later, all rats were killed for serum triglyceride (TG), cholesterol (TC), low density lipoprotein cholesterol (LDL - C), high density lipoprotein cholesterol (HDL - C), levels of TNF -α and arteriae carotis eommunis pathology determination by Immunohistochemistry and Westen - blot methods. Results: Compared with the model group and the aspirin group, the levels of serum TG, TC and LDL - C in dioscornin high dosage group were significantly decreased. However, there was no significant difference between dioscornin low dosage group and model groups. The levels of serum HDL - C were no significantly differences among all groups. In the impaired side, W/L ratio of carotid artery in the model group was thicker obviously than that in sham operation group. Meanwhile there was plaque formed locally. The W/L ratios of carotid artery in dioscornin low and high dosage group were thinner than that in model group. That of dioscornin high dosage group was thinner than aspirin group. The W/L ratios of carotid artery in dioscornin low dosage group and model group were no significant difference. Immunohistochemistry staining and Western blot method showed that the levels of TNF - α in dioscornin low and high dosage groups were significantly decreased compared with the model group. That of dioscornin high dosage was significantly lower than that of aspirin group. That in dioscornin low dosage group and aspirin group were no significant difference.
