摘要 通过比较磷脂酶A1(Lecitase Ultra)(PLA1)和胰脂酶(pancreatic lipase,PL)水解大豆油释放出的脂肪酸组成,推断PLA1催化水解甘油三酯(TAG)具有Sn-1,3位立体选择性。测定了PLA1催化水解大豆油的水解平衡常数。通过分析不同水解时间水解产物的变化,推断反应初期TAG按照1,2-DAG(甘油二酯)水解路线水解,DAG中以1,2-DAG含量为主,随着1,2-DAG含量增加,水解同时按1,3-DAG路线进行,1,3-DAG含量开始增加。在反应后期由于1,3-DAG更容易水解,1,3-DAG浓度逐渐下降,水解反应以1,3-DAG路线水解为主。 Phospholipase A1 (Lecitase Ultra) (PLAI) was deduced to possess Sn - 1,3 - stereoselectivity to triacylglycerol by comparing the composition of the released fatty acid by PLA1 with that by pancreatic lipase during partial hydrolysis of soybean oil. The equilibrium constant of partial hydrolysis of soybean oil with PLA1 was determined. By analyzing the composition of hydrolysate, it was deduced that at the initial stage, triacylglycerol (TAG) was hydrolyzed according to the hydrolysis route of 1,2 -diacylglycerol ( DAG), and the major portion of DAG was 1,2 - DAG. Along with the increase of 1,2 - DAG content, TAG was hydrolyzed according to the hydrolysis route of 1,3 - DAG at the same time. At the last stage, TAG was mainly hydrolyzed by route of 1,3 - DAG and the concentration of 1,3 - DAG decreased gradually because 1,3 - DAG was hydrolyzed more easily.
