摘要
采用细胞增殖试验(MTT法)、Annexin-V/PI双重染色法检测镰形棘豆提取物对体外培养的肝癌细胞增殖抑制及凋亡的影响。结果显示,镰形棘豆提取物可显著抑制人肝癌细胞的增殖活性,尤其当浓度为250μg/mL和500μg/mL时,对肝癌细胞Smmc-7721抑制率均超过了60%,并且高剂量组中处于凋亡早期癌细胞的比率显著高于对照组(P<0.05)。结果表明,镰形棘豆提取物对癌细胞增殖有显著的抑制作用,其作用机制可能与其抑制癌细胞增殖活性和诱导癌细胞凋亡有关。
To explore the proliferative and apoptotic effects of Tibet Medicine Oxytropis falcate extracts on human liver cancer (Smmc-7721 cells),MTT assay and Annexin-V/PI double-staining assay were used to evaluate the effects of proliferation inhibitory and determine the cell apoptosis rate of liver cancer cells in vitro. Compared with the control group, the proliferation inhibitory rates were increased obviously, especially when the Smmc-7721 cells treated with 250 μg/mL and 500 μg/mL Oxytropis falcate extracts, the inhibitory rate is overcome 60%. And the apoptotic rates were increased markedly (P 〈0.05). The extracts of Oxytropis falcate could inhibit the growth of tumor cells in vitro effectively. It may be relative to inhibiting the proliferation activity and inducing the cancer cell apoptosis.
