摘要
目的探讨冬虫夏草对马兜铃酸肾病(aristolochic acid nephropathy,AAN)大鼠模型肾纤维化的保护作用。方法将30只SD大鼠随机分为正常组、模型组和治疗组,每组均为10只。治疗组给予冬虫夏草进行干预,30d后比较各组大鼠肾功能,并通过Masson染色观察各组大鼠肾脏病理变化,RT-PCR法测定各组大鼠肾组织Ⅰ型胶原(collagenⅠ)mRNA的表达。结果模型组大鼠肾脏明显纤维化,其collagenⅠmRNA的表达亦显著升高。冬虫夏草可下调AAN大鼠BUN、SCr水平及col-lagenⅠmRNA的表达及肾脏胶原面积百分比。结论冬虫夏草可有效抑制AAN大鼠模型肾纤维化,其机制可能与下调collagenⅠmRNA的表达、阻断胶原沉积相关。
Objective To investigate the effect of Chinese caterpilar fungus on renal interstitial fibrosis of rats with aristolochic acid nephropathy. Methods All SD rats in the study were randomly divided into normal group (n=10) ,model group(n=10)and treatment group(n= 10). The treatment group was administered with Chinese caterpilar fungus for 30 days. The renal function were detected, the pathological changes in the kidneys were observed under light microscope with masson staining. RT-PCR method was employed to detect the expression of collagen Ⅰ mRNA in kidney. Results In the model kidneys,there were significant renal interstieial fibrosis. RT-PCR showed that the expression of collagen Ⅰ mRNA were significantly increased in the model kidneys(P〈0.05). In comparison with the model group, BUN and Scr levels, and the expression of collagen Ⅰ mRNA in the treatment group were significandy decreased(P〈0.05). Conclusion Chinese caterpilar fungus can down-regulate the expression of collagen Ⅰ mRNA in aristolochic acid nephropathy rats, through which Chinese caterpilar fungus might mitigate the pathological changes of the aristolochic acid nephropathy renal tissue.
