摘要
通过对29头德宏水牛的细胞色素b基因(Cytb)全序列的测序和分析,发现Cytb基因全长1 140bp,共检测到了7种单倍型、14个核苷酸的多态位点,单倍型多样度(Hd)为0.672,核苷酸多样度(Pi)为0.002 3,表明德宏水牛具有丰富的遗传多样性。同时发现德宏水牛Cytb基因具有两种终止密码子AGA和AGG。NJ系统发育树构建的结果表明德宏水牛Cytb基因有2个支系A和B,这说明德宏水牛起源于2个母系。
Complete sequences of mtDNA Cytb gene from 29 samples in Dehong buffalo population were sequenced and analyzed. The results showed that the length of the complete mtDNA Cytb sequence was 1 140 bp, and 7 haplotypes with 14 nucleotide polymorphisms were detected. The haplotype diversity and nucleotide diversity were 0.672 and 0.002 3, respectively, indicating abundant genetic diversity in Dehong buffaloes. There were two stop codons of AGA and AGG detected in Cytb gene in Dehong buffaloes. NJ tree constructed by 29 Cytb haplotypes revealed two highly divergent lineages A and B, which suggested that Dehong buffalo had two maternal origins.
