摘要
目的研究尼莫地平(nimodipine,NM)对戊四氮(pentylenetetrazol,PTZ)致痫大鼠海马核转录因子-κB(NF-κB)蛋白表达的影响,探讨尼莫地平的脑保护机制。方法将SD大鼠随机分为正常对照(NC)组,癫痫持续状态(status epilepsy,SE)组和NM组,观察3组大鼠的行为学改变,并用免疫组化方法和Western blot方法测定NF-κB蛋白的表达。结果与SE组相比,NM组癫痫发作级别明显减低,发作潜伏期延长。SE组和NM组大鼠NF-κB阳性细胞数、NF-κB蛋白表达水平均高于NC组,差异有统计学意义(P<0.05)。NM组大鼠NF-κB阳性细胞数、NF-κB蛋白表达水平均低于SE组,差异有统计学意义(P<0.05)。结论NM具有神经保护作用,该作用可能是通过抑制NF-κB蛋白的活化来实现的。
Objective To study the influence of nimodipine(NM) on NF-κB expression in hippocampus of pentylenetetrazol(PTZ) induced epileptic rats,and to discuss the mechanism of neuroprotective effect of NM.Methods SD rats were divided into groups NC(normal control),SE(status epilepsy) and NM.Ethological changes were observed,and the expression of NF-κB were measured by immunohistochemistry and Westernblot.Results Epileptic discharge was significantly decreased and seizure latency prolonged in NM group as compared with SE group.The positive cell count and the expression of NF-KB was significantly higher in groups SE,NM than in NC group,the difference was significant(P〈0.05),and was lower in NM group than in SE group,the difference was significant(P〈0.05).Conclusion NM has a neuroprotective effect against epilepsy by inhibiting the activation of NF-KB proteins.
