摘要
目的探讨脑缺血再灌注大鼠脑组织病理形态的变化以及醒脑开窍针刺方法对脑组织的影响。方法45只SD大鼠随机分为正常组、模型组、假手术组、针刺组和非穴位针刺组,除正常组5只外,其余4组各10只,采用线栓法建立脑缺血再灌注模型,针刺组采用醒脑开窍针法,非穴位针刺组直接针刺非穴位点不行手法,正常组、假手术组和模型组相同时间点抓取,不做其他处理。除正常组外其余4组分设造模后6h和24h两个观察时间点,通过光镜与电镜观察缺血侧大鼠脑组织病理形态的变化。结果模型组大鼠脑组织大量神经元变性、坏死和大量炎细胞浸润等病理损害,针刺组各时间点均较模型组同时间点病变程度明显减轻,非穴位针刺组各时间点病变程度依然随时间段的延长而渐趋加重,和同时间点模型组相比并无明显改善。结论醒脑开窍针法可使不同缺血时相脑组织损伤得到改善,对临床治疗脑缺血具有重要的意义。
Objective To explore the pathomorphological changes of brain tissues of rats with ischemic-reperfusion damage and observe the effect of resuscitating acupuncture therapy on the brain tissues. Methods Totally 45 SD rats were randomized into normal group, model group, pseudo operation group, acupuncture group and non-point acupuncture group. The normal group had 5 rats in it and the rest 4 groups had 10 rats in each. Thread-blocking method was used to make the models of cerebral ischemic-reperfusion damage in the 4 groups. The acupuncture group was treated with resuscitating acupuncture therapy, the non-point acupuncture group with acupuncture at non-points and without manipulation of needles, the normal group, model group, and pseudo operation group was caught and released without any other treatment. The observation time point was at 6h and 24h after modeling for all the groups except the normal group. Pathomorphological changes of brain tissues on the ischemic side were observed with optical and electron microscopes. Results In the model group, there were pathomorphological changes of neuronal degeneration, necrosis and massive inflammatory cell infiltration; in the acupuncture group, pathomorphological changes were mild in each time point than those in the model group; in the non-point acupuncture group, pathomorphological changes were getting worse with the time going on, without much improvement as compared with the same time point in the model group. Conclusion Resuscitating acupuncture therapy could improve the pathological condition in different time phases of brain tissues with ischemic-reperfusion damage in rats, being significant in clinical treatment for cerebral ischemia.
