摘要
采用浸渗法制备萝卜种子抗真菌蛋白Rs-AFPs粗提液进行抑菌试验,结果表明:在萝卜种子发育过程中,Rs-AFPs是在一定时期(约开花后45d)开始合成并逐步积累的,因此充分成熟的萝卜种子Rs-AFPs粗提液具有较强的抑菌活性。Rs-AFPs的耐酸性、耐碱性、热稳定性及对不同病原真菌的抗性研究结果表明,在pH为4和10时.Rs-AFPs能保持90%的活性;在80℃水浴60min其活性仍为100%,在100℃水浴30min和45min,分别具有80%和50%的活性;在4℃下存放5、20、35、45d,Rs-AFPs的活性分别保持100%、90%、80%和70%。Rs-AFPs对植物丝状病原真菌具有广谱抗性,如对供试病原菌镰刀菌、疫霉菌、芭蕉炭疽菌和稻瘟菌均表现出颇强的抑菌活性,Rs-AFPs无抗细菌活性。
The biological properties of Raphanus sativus-antifungal proteins (Rs - AFPs )was studied by using radish seed inhibition solution. Rs -AFPs extracts were obtained from radish seeds at various development stages. Results demonstrated that Rs -AFPs began to be synthesized at a certain seed development stage (at some 45 days after anthesis ) and accumulated gradually and that Rs - AFPs extracts from fully mature radish seeds exhibited higher antifungal activity than those from immature ones. The stability of antifungal activity of Rs-AFPs under acid and alkaline and heat conditions was determined. Results disclosed that Rs - AFPs maintained 90% of their activity under pH4 and pH10. The Rs -AFPs extracts kept at 80℃ for 60 min were found to have 100% activity, and at 100℃ for 30 min and 45 min 80% and 50% respectively. The extracts stored at 4℃ for 5,20,35 and 45 days still had an activity of 100%,90%,80% and 70% respectively.Rs -AFPs exhibited potent antifungal activity against a broad spectrum of filamentous pathogenic fungi of plants,such as Colletotrichum musae,Piricularia oryzae,Fusarium oxysorum and Phytophthora citriophthora. But Rs -AFPs was not resistant to bacteria.
